It looks like the COM port of your cellular modem may have changed or the hardware is experiencing some sort of issue.

To check the COM port first, go in to the 'Phone and Modem' section of the Control Panel in Windows. Once there, go under the Modems tab. What's listed and what COM port(s) are shown?

I'd first recommend removing the devices that aren't present. Not that you have to do that, it's just better to keep a tidy hardware list.

Next, you'll need to go in to the properties of the device attached to COM21, then go under the Advanced tab. Once there, click Advanced Port Settings and specify a COM port number 16 or lower.

After specifying the new COM port, update the connector to use it and that should fix the issue.

If you would, try to run Putty, Hyperterminal or some other port testing utility.

Then connect to COM2 with 115200 baud rate, 8 data bits, 1 stop bit and no parity.

Are you able to establish a connection to the port?

If so, what happens if you type any of the following and hit enter?
AT
AT+CSQ

Hm... if it can't establish a connection, I'd recommend contacting the cellular modem's manufacturer for assistance as it appears that the device is malfunctioning and can't be reached.
